The Impact of tRNA-Phe Anticodon Loop Modifications on Oxidative Stress Recovery in Saccharomyces cerevisiae

Abstract

"Transfer RNA anticodon loop modifications in the form of 2`-O methylations are critical for efficient translation in Saccharomyces cerevisiae (brewer’s yeast). The methyltransferase Trm7 associates with proteins Trm732 and Trm734 to methylate the thirty-second (C-32) and thirtyfourth (G- 34) nucleotides of tRNA- Phe, respectively. Yeast strains lacking either modification show healthy phenotypes, but sick growth is exhibited when both modifications are absent. Because oxidative stress necessitates efficient protein production, translational deficiencies may be magnified in trm732Δ and trm734Δ exposed to oxidative stress. It is established that trm7Δ mutants show lowered oxidative stress resistance, but the relative extent of trm732Δ and trm734Δ stress resistance is less understood. Spot tests confirm lowered stress resistance in trm7Δ mutants, but not in trm732Δ and trm734Δ mutants."
